Biologists are very interested to see what part of the spectrum is absorbed by various leaves (photosynthesis).
Here, we’ll do a quick setup which allows us to measure this absorbance using the Jaz spectrometer and a reference lightsource.
This setup has the following:
- Jaz Spectrometer
- Jaz Battery Pack
- Collimating lens 74-UV DA (Direct Attach) or 74-UV with fiber
- WS1 Reflection standard
And, here’s the experiment:
- Connect the 74-UV.
- Set up the integrating time of the spectrometer measureing the sunlight via the WS1 reflection standard. Hold Jaz at a 45 degree angle to do this.
- Set up the boxcar to get some smoothing of the spectrum.
- Go into the right measurement mode.
- Perform the dark calibration and reference calibation at a 45 degree angle.
- Find some leaves to perform the measurements with and store the results.
Questions:
- What should we use as a reference lightsource?
- What kind of measurement are we going to perform — absorption or reflection?
- Do you see anything special in the spectra you measured?
